GMC0403注射液对氰化钠诱导原代培养心肌细胞损伤的保护作用

摘要 目的研究GMC0403注射液对氰化钠诱导原代培养心肌细胞损伤的保护作用。方法采用1μmol/ml氰化钠与低糖DMEM、共培养复制心肌细胞缺糖、缺氧损伤模型,通过细胞线粒体琥珀酸脱氢酶活力(MSDH)、乳酸脱氢酶(LDH)外漏率、台盼蓝染色率研究GMC0403注射液对原代培养心肌细胞氰化钠(NaCN)诱导缺糖、缺氧模型的保护作用。结果GMC0403注射液能显著降低缺糖、缺氧损伤心肌细胞线粒体琥珀酸脱氢酶活力,抑制LDH的外漏,降低台盼蓝染色率。结论GMC0403注射液对NaCN诱导原代培养心肌细胞损伤具有保护作用。 Objective: To investigate protective effects of GMC0403 injection on primary culture of rat cardiac myocyte injury induced by NaCN in vitro. Methods:After adding GMC0403 injection, the primary culture of rat cardiac myocyte was injured by 1μmol/ml NaCN and low glucose DMEM. The release ratio of lactic dehydrogenase (LDH) from cardiac myocyte, mitocbondrion succinic dehydrogenase (MSDH), and staining ratio by trypan-blue were assayed. Results: GMC0403 injection significantly increased the MSDH activity (volume of A570 significantly increasing), decreased the release ratio and staining ratio of trypan-blue. Conclusion: GMC0403 injection could ameliorate the primary culture of cardiac myocyte induced by NaCN.
