摘要 目的研究硒化壳聚糖对NB4细胞第二信使的影响,探讨其抑制NB4细胞生长,诱导其凋亡的机制。方法流式细胞法观察药物对细胞内钙离子的影响;放免法检测药物对细胞内cAMP和cGMP含量的影响。结果硒化壳聚糖作用24h,NB4细胞内钙离子和cAMP含量明显升高,cGMP含量明显降低,cAMP/cGMP比值增大。结论硒化壳聚糖可通过升高细胞钙离子和cAMP含量,降低cGMP含量,增大cAMP/cGMP比值来抑制NB4细胞生长,诱导其凋亡。 Objective:TO Study the effects of selenium chitosan on the level of Ca^2+,cAMP and cGMP in NB4 cells and to explore its possible mechanisms in inducing apoptosis and inhibiting proliferation. Methods : The change of Ca^2+ content was observed by using flow cytometry; radioimmunity method was used to determine the change of cAMP and cGMP content. Results: After incubation for 24h with 50-200mg L^-1 of selenium chitosan ,the levels of Ca^2+ and cAMP increased significantly (P 〈0. 01) and the level of cGMP decreased evidently (P〈0.01)as compared with those of the controls. Conclusions: Selenium chitosan could reduce cGMP level, promote Ca^2+ and cAMP level and cAMP/ cGMP ratio, which are all possible mechanisms that selenium chitosan induces NB4 cell apoptosis and inhibite proliferation.
