摘要
目的:探讨兔眼局部应用盐酸川芎嗪滴眼液后前房穿透性。方法:18只白兔随机分为6组,并于各组白兔结膜囊内滴入50μL盐酸川芎嗪滴眼液,分别于5、15、30、60、120、180min取房水,采用高效液相色谱法进行测定。色谱柱为DiamonsilCl8不锈钢柱(250mm×4.6mm,5μm);流动相为甲醇:水=62:38;流速:0.9mL/min;检测波长为280nm。结果:5、15、30、60、120、180min各组的房水浓度分别为(15.785±2.988)μg/mL,(11.900±1.743)μg/mL,(8.286±1.182)μg/mL,(2.745±0.807)μg/mL,(0.379±0.091)μg/mL,(0.049±0.038)μg/mL,其中5min为最高峰,然后逐渐下降,180min后房水浓度已降到很低。结论:盐酸川芎嗪滴眼液滴眼后房水穿透性良好,这一结果为盐酸川芎嗪滴眼液局部应用后治疗眼前部疾病提供了实验依据。
Purpose: To study the aqueous humor penetration of ligustrazine hydrochloride after ocular administration in rabbits. Methods: Eighteen rabbits were randomly divided into 6 groups. The aqueous humor was collected at 5,15,30,60,120 and 180 minutes following topical administration of ligustrazine hydrochloride eye drops and the concentration of ligustrazine hydrochloride was determined by HPLC. Analytical column was Diamonsil C18 stainless steel column (250 mm ×4.6 mm, 5μm);The mobile phase was methanol: water(62:38); Flow rate was 0.9 ml/min;The detection wavelength was 280 nm. Results: The concentrations of ligustrazine hydrochloride in aqueous humor were ( 15.785 ±2.988 ) μg/mL, ( 11.900 ± 1.743 ) μg/mL, ( 8.286 ± 1.182) μg/mL, (2.745 ± 0.807) μg/mL, (0.379±0.091) μg/mL, (0.049±0.038) μg/mL, respectively, after a time interval of 5 min, 15 min, 30 min, 60 min, 120 min, 180 min. The maximum concentration was 15.785 μg/mL at 5 min, and then the concerntration of ligustrazine hydrochloride gradually decreased and declined to a lower point at 180 min.Conclusion: The penetration of ocular administration was good. management of eye diseases with ligustrazine hydrochloride in aqueous humor after The results provided experimental data for the ligustrazine hydrochloride by topical administration.
