摘要
目的从蛋白质组学角度探讨偏头痛肝阳上亢证证候相关蛋白及快速埋线缓解疼痛和改善肝阳上亢证症状的机制。方法用固相pH梯度双向凝胶电泳分离正常大鼠及肝阳上亢证大鼠经快速埋线治疗前、后肾上腺的总蛋白质,经考马斯亮兰染色,PDQuest 7.0软件分析。结果与正常组匹配,肝阳上亢证组与快速埋线组大鼠肾上腺图象匹配率分别为85%、89%,且重复性较好,其蛋白质主要分布在等电点PI3-8,分子量M r14.4-75kD范围。与正常组比较,肝阳上亢证组中蛋白质点表达上升2倍及以上的点有10个,下降2倍及以上的点有12个。而在快速埋线组中这些表达上升的点均有下降,有11个点下降达2倍以上,其中有7个下降后接近于正常组;而表达下降的点均有上升,有8个点上升达2倍以上,其中有5个点上升后接近于正常组。结论这些有显著差异的蛋白质可能是偏头痛肝阳上亢证证候相关蛋白;快速埋线缓解头痛和改善临床症状也可能与上述蛋白质的改变有关。
Objective To explore the protein associated Gan-Yang-Shang-Kang syndrome (CYSKS) from proteome, and discuss the mechanism of alleviating headache and ameliorating symptom of rapid embedding thread therapy on CYSKS of migraine rats. Methods The total proteins of adrenal glands of normal control rats and CYSKS of migraine rats were separated by immobilized pH gradient - based two - dimensional gel electrophoresis (2 - DE). After coomassie blue staining, PDQuest 7.0 software was applied to analyze the 2 - DE images. Results Compared with normal control group, the average matching rates of adrenal glands of CYSKS group and treatment group were 85% and 89% with reproducibility, respectively. Proteins were mainly displayed at pI 3 -8, Mr 14. 4 -75kD range. At CYSKS rats, 10 spots expressed two times than those of normal rats, 12 spots expressed half than those of normal rats. After treatment, 11 spots of highly expressed proteins decreased, 7 spots of them were near to those of normal rats. 8 spots of depressed proteins increased, 5 spots of them were near to those of normal rats. Conclusions These protein spots with significant difference may be associated with Gan-Yang-Shang-Kang syndrome (CYSKS) in migraine. After treatment by rapid embedding thread therapy, the alleviation of headache and the amelioration of symptom of CYSKS may be relative with some of these proteins spots.
