摘要
目的探讨β-连接素(β-catenin)和γ-连接素(γ-catenin)基因在鼻咽癌及其淋巴结转移中的表达及意义。方法应用免疫组化SP法检测15例鼻咽黏膜慢性炎、28例分化型非角化性鼻咽癌和20例鼻咽癌淋巴结转移中β-catenin和γ-catenin的表达。结果β-catenin、γ-catenin在分化型非角化性鼻咽癌及其淋巴结转移中的异常表达率均分别为85.7%(24/28)和95.0%(19/20),均显著高于鼻咽黏膜慢性炎[分别为13.3%(2/15)和6.7%(1/15)](P〈0.01)。β-catenin、γ-catenin在鼻咽黏膜慢性炎中的染色分级分别为2.80±0.56和2.93±0.26;分化型非角化性鼻咽癌为0.79±1.07和0.86±1.15;鼻咽癌淋巴结转移为0.25±0.72和0.30±0.73;三组别中两两比较差异有统计学意义(P〈0.05)。β-catenin和γ-catenin在鼻咽黏膜慢性炎、分化型非角化性鼻咽癌及其淋巴结转移中均呈正相关关系(rs=0.89,0.61,0.94,P〈0.01)。结论β-catenin、γ-catenin基因异常表达可能共同参与鼻咽癌的侵袭和转移;β-catenin和γ-catenin染色分级可能为判断鼻咽癌转移的更敏感指标。
Objective To investigate the expressions of β-catenin and γ-catenin gene in differentiated non - keratinizing nasopharyngeal carcinoma (NPC) and lymph nodes with NPC metastasis, and their biological significance. Methods Expressions of β-catenin and γ-catenin were detected by immunohistochemical method in 15 patients with chronic nasopharyngitis, 28 patients with differentiated non - keratinizing carcinoma and 20 patients with lymphy node metastasis of the NPC. Results In differentiated non - keratinizing carcinoma and lymph node metastasis of the NPC, the abnormal expression rates of β-catenin and γ-catenin were 85.7% ( 24/28 ) and 95.0% (19/20) respectively, which were significantly higher than that of chronic nasopharyngitis( 13.3% ,2/15 and 6.7%, 1/15)respectively)( P 〈0. 01 ). In differentiated non - keratinizing carcinoma and lymph node metastasis of the NPC, the staining grades of β-catenin and γ-catenin were 2. 80 0. 56 and 2. 93 ± 0. 26 in chronic nasopharyngitis, 0. 79 ± 1.07 and 0. 86 ± 1.15 in differentiated non-keratinizing carcinoma and 0. 25 ± 0. 72 and 0. 30 ± 0. 73 in lymph node metastasis of the NPC, respectively. The staining grades of β-catenin and β-catenin among the three groups had significant differences( P 〈0. 01 or P 〈0. 05 ). The expression of β-catenin and β-catenin showed a positive correlation( P 〈 0. 01 ) in chronic nasopharyngitis, differentiated non - keratinizing carcinoma and lymph node metastasis of the NPC. Conclusions The abnormal expression of [^-catenin and β-catenin may together participate in the invasion and metastasis of NPC. The staining grades of β- catenin and β-catenin may be more sensitive at predicting metastasis of NPC.
