摘要
目的研究自然状态下乙型肝炎病毒(HBV)基因多态性及其对病毒复制能力的影响。方法用基因芯片技术检测157例未经抗病毒治疗的乙型肝炎(乙肝)患者HBV基因组前C区(1896位、1814位)、基本C区启动子(1762位、1764位)、P区(204位、180位)基因变异发生情况,同时检测其HBV DNA载量。结果自然状态下,HBV各位点累计自发变异率为50.96%,前C区、BCP和P区的单独变异率分别为9.55%、31.85%和1.91%,前C区和BCP联合变异率为7.64%。变异组HBV DNA载量(lg值)为6.14±1.35,野生组为5.89±1.37(P>0.05)。结论自然状态下HBV基因前C区、BCP有很高的变异率,P区的变异率很低,HBV变异并没有改变复制能力。
To study the effect of genetic polymorphism of hepatitis B virus on its replicated potentia in raw state.Methods In 157 non-antiviral therapy patients,we detected the genovariation of pro-C domain(1896,1814),promoter of basi-C domain(1762,1764)and P domain(204,180)of HBV genome with gene micro array technique,and the HBV DNA quantum with fluorescent quantitation PCR.Results In raw state,spontaneous variation of HBV was 50.96%.The aberration rates of pro-C domain,basi-C domain and P domain were 9.55%,31.85% and 1.91%,respectively.The aberration rate of pro-C domain combined with basi-C domain was 7.64%.HBV DNA quantum of the variation group(log value)was 6.14±1.35,and of grow wild group 5.89±1.37(P〉0.05).Conclusion In raw state,aberration rate in pro-C
domain and basi-C domain of HBV genome is higher than that in P domain,and variation of HBV does not influence its replicated potentia.
