VEGF-C表达对胰腺癌细胞凋亡的影响 被引量:1

摘要 目的研究VEGF-C反义寡核苷酸对胰腺癌原位种植瘤模型中PANC-1细胞凋亡的影响。方法建立人胰腺癌细胞株PANC-1裸鼠原位种植瘤自发性淋巴结转移模型,分离、原代培养原位灶和淋巴结转移灶中的胰腺癌细胞,并应用realtime定量PCR、流式细胞术检测VEGF-C反义寡核苷酸转染对其凋亡能力的影响。结果VEGF-C反义寡核苷酸抑制胰腺癌细胞VEGF-C的表达后,淋巴结转移灶组PANC-1细胞凋亡率为(15.2±2.7)%,显著高于空白组、SODN组(P<0.05);原发灶组PANC-1细胞凋亡率为(5.5±2.7)%,与空白组、SODN组的差异无统计学意义(P>0.05);并且淋巴结转移灶组PANC-1细胞的凋亡率高于原发灶组,差异有统计学意义(P<0.05)。结论VEGF-C表达下调能显著诱导淋巴结转移灶中胰腺癌细胞的凋亡。 Objective To investigate the effect of antisense oligonucleotide(ASODN) of vascular endothelial growth factor C(VEGF-C) on apoptosis of PANC-1 cells in pancreatic cancer.Methods The spontaneous lymphatic metastasis model in nude mice with orthotopic implantation for the human pancreatic cancer cell line PANC-1 was established.Pancreatic cancer cells of primary tumor and spontaneous lymphatic metastasis were isolated and cultured,and divided into control group,ASODN group and scramble oligonucleotide(SODN) group.The effect of VEGF-C ASODN on apoptosis of pancreatic cancer cells were detected by real-time quantitative polymerase chain reaction(PCR) and flow cytometry.Results VEGF-C ASODN inhibited the expression of VEGF-C in pancreatic cancer cells.The apoptosis rate of pancreatic cancer cell derived from lymphatic metastasis was(15.2±2.7)%,higher than in control group and SODN group significantly(P〈0.05);while the apoptosis rate of pancreatic cancer cell derived from primary tumor was(5.5±2.7)%,but not significantly different from control group and SODN group(P〉0.05).And the apoptosis rate of pancreatic cancer cell derived from lymphatic metastasis was significantly higher than from primary tumor(P〈0.05).Conclusion Apoptosis of pancreatic cancer cells derived from spontaneous lymphatic metastasis could be induced by inhibition of VEGF-C,but not from primary tumor.
