摘要 目的探讨沙眼衣原体细胞增殖培养和鉴定的方法。方法将沙眼衣原体接种于Hep-2细胞中,收获培养物,将其分别用Giemsa染色、免疫层析法、PCR技术和核苷酸序列检测方法进行鉴定。结果经免疫层析法鉴定所收获的培养物为沙眼衣原体,并经Giemsa染色,可见其典型包涵体结构,PCR技术和核苷酸序列检测方法进一步鉴定其血清型别为D型。结论Hep-2细胞可用于沙眼衣原体的增殖培养。 Objective To cultivate chlamydia trachomatis and identify it. Methods Chlamydia trachomatis was cultivated in Hep - 2 cell monolayer and identified by Giemsa staining method, immunochromatographic technique, PCR analysis and DNA sequencing. Results Chlamydia trachomatis was detected by immunochromatographic technique, and the inclusion could be identified by Giemsa staining method. The serotype of chlamydia trachomatis was D type identified by PCR analysis and DNA sequencing. Conclusion Chlamydia trachomatis could be cultivated in the Hep - 2 cell.
