摘要
初步提纯的马铃薯酷氨酸酶(TYR)固定于海藻酸钠/二氧化硅(ALG/SiO2)凝胶复合物中,并用于含酚废水的处理。在pH=7.0时,固定化酪氨酸酶的比活力为1120 U/g。25℃,以邻苯二酚作底物时,其米氏常数为10.6 mmol/L。相对于游离酶和海藻酸钠固定化酶,本杂化凝胶固定酷氨酸酶具有良好的热稳定性、贮存稳定性及连续使用性。研究表明,本固定化酪氨酸酶有可能实际应用于含酚废水的处理。
An organic-inorganic hybrid gel of alginate/SiO2 ( ALG/SiO2 ) was used for immobilizing the partially purified potato tyrosinase(TYR) for the treatment of phenohc wastewater. The highest activity of immobihzed TYR was 1 120 U/g at pH 7. 0. The Michaelis constant for immobihzed PPO was 10. 6 mmol/L at 25℃ when catechol was used as substrate. This immobilized TYR exhibited a higher thermal stability, storage stability and comtinuous use stabihty than that of free TYR and alginate-immobihzed TYR. This study shows that alginate/SiO2-immobihzed TYR might be a practical tool for removal of phenohc compounds from industrial wastewater.
