摘要
目的:观察三七总皂苷(PNS)对小鼠巨噬细胞源性泡沫细胞形成的影响,以探讨PNS抗动脉粥样硬化(AS)作用的机制。方法:采用小鼠腹腔巨噬细胞与40mg.L-1的氧化低密度脂蛋白(ox-LDL)孵育72h建立泡沫细胞模型,用PNS进行干预。油红O染色观察细胞内脂质堆积情况,酶法测定细胞内总胆固醇(TC)、游离胆固醇(FC)、胆固醇酯(CE)的含量及CE/TC比值。结果:模型组细胞中TC、FC、CE及CE/TC均显著高于对照组细胞(P<0.01);与模型组比较,PNS高、中剂量组细胞内TC、FC、CE及CE/TC均有显著性降低(P<0.01);且PNS各剂量组间有显著性差异。形态学上,PNS高、中剂量组细胞的油红O染色阳性细胞数显著减少。结论:PNS可能通过抑制泡沫细胞的形成发挥抗AS的作用。
OBJECTIVE: To observe the effect of total Saponins of Panax Notoginseng (PNS) on the formation of macrophage- derived foam cells and explore its antiatherogenic mechanism. METHODS: In order to examine the effect of PNS, macrophage cells from mouse peritoneal were incubated with 40m·L^-1 ox- LDL for 72h to establish foam cell model. The cellular lipid accumulation was examined by oil red staining. The cellular contents of total cholesterol (TC), free cholesterol (FC) and cholesterol ester (CE) and the ratio of CE/TC were detected by enzymic method. RESULTS: The contents of TC, FC, CE (cholesteryl ester) and CE/TC ratio in model group were significantly higher than in control group(P〈0.01) . Compared with model group, the contents of TC, FC, CE and CE/ TC ratio were significantly reduced in high - dose and medium- dose PNS subgroups (P (0.01), and there were significant differences among PNS subgroups. Morphologically, the number of oil red - positive macrophages was decreased significantly in high - dose and medium - dose PNS subgroups. CON- CLUSION: PNS might exert its antiatherogenic effect through inhibiting the formation of macrophage - derived foam cells.
