摘要
目的观察黑米花色苷及与化疗药物联合对人乳腺癌MCF-7细胞株及白血病HL-60细胞株增殖的影响。方法MTT法检测黑米花色苷及与紫杉醇(paclitaxel,PXT)、阿霉素(doxorubicin,DOX)联合,对MCF-7及HL-60细胞增殖的影响;流式细胞仪分析黑米花色苷及与DOX联合,对HL-60细胞周期分布及凋亡的影响。结果黑米花色苷对MCF-7细胞增殖无显著作用,但对HL-60细胞具有明显的增殖抑制作用,能够有效诱导HL-60细胞凋亡,并引起G0/G1期阻滞;同时,黑米花色苷与PXT联合作用MCF-7和HL-60细胞,以及与DOX联合作用MCF-7细胞,均无显著拮抗或协同作用;黑米花色苷与低剂量DOX联合作用HL-60细胞具有显著协同作用,而黑米花色苷与较高剂量DOX联合作用HL-60细胞则具有显著拮抗作用,能够显著抑制DOX引起的细胞凋亡。结论黑米花色苷对HL-60细胞具有显著增殖抑制作用,与低剂量DOX联合具有协同作用,与较高剂量DOX联合则具有拮抗作用。
Objective To observe the effects of anthocyanin-rich extract from black rice alone or combined with chemotherapeutic agents on proliferation of human breast cancer cell line MCF-7 and leukemia cell line HL-60. Methods The proliferation of MCF-7 and HL-60 cells was measured by MTF assay. The distribution of cell cycle and apoptosis of HL-60 were determined by flow cytometry. Results Anthocyanin-rich extract from black rice could inhibit the proliferation of HL-60 and induce apoptosis and G0/G1 arrest, but it had no significant effect on MCF-7. The combined effects of anthocyanin-rich extract with paclitaxel or doxorubicin on MCF-7 and paclitaxel on HL-60 were additive. The combined effects of anthocyanin-rich extract with low-dose doxorubicin on HL-60 were synergetic and that with high-dose doxorubicin were antagonistic. Conclusion Anthocyanin-rich extract from black rice could significantly inhibit the proliferation of HL-60.
