摘要
目的观察骨桥蛋白(OPN)在食管鳞癌组织中的表达状况,同时观察食管鳞癌组织中血管生成状况,探讨OPN与血管生成的关系。方法运用免疫组化S-P法做44例食管鳞癌组织兔抗人骨桥蛋白抗体染色,用抗CD34单克隆抗体做44例食管鳞癌组织免疫组化染色,并在显微镜下作MVD计数。结果OPN在食管癌组织的表达率为86.3%。癌组织中OPN主要表达于肿瘤细胞的细胞浆中。全组44例标本MVD计数为24.07±5.82,癌巢及癌细胞密集区MVD少于癌周区域。本研究提示,OPN阳性组MVD高于阴性组(P<0.05)。等级相关分析,结果表明OPN表达与MVD呈正相关(r=0.320,P=0.034)。结论食管鳞癌组织中OPN主要由肿瘤细胞产生。食管鳞癌中存在活跃的血管生成。OPN与血管生成关系密切。
Objective To observe the expression of osteopontin(OPN) and the expression of CD34 and count microvessel density in esophageal squamons cell carcinoma, to explore the effect of OPN in tumor anglogenesis. Methods S-P inanunohistochemical staining was used to investigate the expression of OPN by anti-osteopontin polyclonal antibody and the expression of CD34 by anti-CD34 polyclonal antibody in the specimens of 44 cases of esophageal squamons cell carcinoma, and nficrovessel density was counted in the high power field ( × 400) of microscope. Results The total positive rate of OPN expression in 44 cases of esophageal squanaous cell carcinoma is 86.3 %. OPN was mainly distributed in cytoplasm of tumor cells. The microvessel density counting of 44 cases of esophageal squamous cell carcinoma is 24.07±5.82. The microvessel of carcinoma nests was less than the area around it. It was found that the MVD of OPN expression in positive was higher than that in negative. Rank correlated analysis showed that OPN and MVD were positive correlated. Conclusion OPN was mainly produced by tumor cells. There was active angiogenesis in esophageal squanaous cell carcinoma. OPN had close relationship with tumor angiogenesis in esophageal squanaons cell carcinoma.
