摘要
目的观察冻融小鼠囊胚的超微结构改变情况,研究冷冻对囊胚的冷冻损伤。方法采用6组玻璃化溶液对小鼠囊胚进行冷冻保存,复苏后通过透射电镜观察超微结构改变情况。结果各组小鼠囊胚均表现相似的超微结构损伤:线粒体肿胀、粗面内质网脱颗粒、高尔基复合体扩张、含有絮状物或膜性结构的小泡增多、微丝结晶的出现、细胞间紧密连接和桥粒减少。结论冷冻对于小鼠囊胚的冷冻损伤主要是细胞膜性结构和细胞骨架系统的损伤。
Objective: To study the cryoinjury by observing the ultrastructural changes of frozen-thawed mouse blastocysts. Methods: Mouse blastocysts were frozen in six kinds of vitrification solutions. After thawing, the ultrastructural changes were observed with transmission electron microscopy. Results: There were similar ultrastructural characteristics in the blastocysts of each group: swollen mitochondria, granule loss of rough endoplasmic reticulum, dilatation of Golgi complexes, increase of vesi- cles containing flock material or membrane residues,appearance of microfilament crystallization, and loss of intercellular tight junctions and desmosomes. Conclusion: The cryoinjury to mouse blastocysts was mostly the damage of cellular membrane and cytoskeletal structure.
