摘要 [目的]探讨Lumican的表达与结直肠癌发生、发展的关系。[方法]构建100对结直肠癌及相应癌旁正常黏膜上皮组织的组织芯片,应用免疫组化方法检测Lumican的表达强度。[结果]Lumican在结直肠癌组织中的表达率(84.0%)显著高于癌旁正常黏膜中的表达率(42.6%)(P<0.01)。Lumican在腺癌中的表达率(87.2%)高于在黏液腺癌及印戒细胞癌中的表达率(68.8%)(P<0.05)。Lumican的表达强度与结直肠癌浸润深度之间存在非常显著的正相关(r=0.07959,P<0.01)及线性关系(χ2=5.2148,P<0.05),即随着浸润程度加深,Lumican的表达强度逐渐增加。Lumican的表达强度与患者的年龄、性别、发病部位、淋巴结是否转移及分化程度无关(P>0.05)。[结论]Lumican的异常表达可能是结直肠癌发生的早期事件,与结直肠癌的组织分型和侵袭能力有关,在结直肠癌的发生发展中发挥重要作用。 [Purpose ]To explore the relationship of Lumican expression with carcinogenesis and progression in coloreetal cancer. [Methods ] One hundred couples of tissue chips of eolorectal cancer tissues and adjacent normal epithelium mucosa tissues were constructed. Expression intensity of Lumincan was measured by immunohistoehemical method. [Results] Lumican expression ratio in colorectal cancer tissues (84.0%) was significantly higher than that (42.6%) in adjacent normal epithelium mucosa tissues (P〈0.01). Lumican expression ratio in adenocareinoma (87.2%) was higher than that in mucinous adenocareinoma and signet-ring cell carcinoma (68.8%), (P〈0.05). There was a distinctly positive correlation between intensity of Lumican expression and invasive depth in colorectal cancer (r=0.07959,P〈0.01), as well as linear correlation (X^2=5.2148,P〈0.05). Lumiean expression intensity increased as invasive depth increased in colorectal cancer. Lumiean expression intensity didn't relate to age, sex, tumor location, lymph node metastasis and cell differentiation (P〉0.05). [Conclusion ] Mal-expression of Lumican correlates to histopathologic type and invasive capability in colorectal cancer. It may be the early event of colorectal carcinogenesis.
