N-甲酰-L-甲硫氨酰-L-亮氨酰-L-苯丙氨酸(fMLP)三肽的合成

摘要 目的合成N-甲酰-L-甲硫氨酰-L-亮氨酰-L-苯丙氨酸三肽(fMLP)。方法以L-亮氨酸、L-苯丙氨酸为原料,采用环酸酐法(NCA)合成二肽L-亮氨酰-L-苯丙氨酸;又以DCC、HOBT为偶联剂,与N-甲酰-L-甲硫氨酸进行偶联,在0.5mol.L-1的NaOH溶液中水解得到fMLP三肽。结果与结论合成的三肽fMLP总收率为44.0%。其结构经IR、1H-NMR、MS确证。 Aim To study the synthesis of tripeptide fMLP. Methods L-Leu-L-Phe was synthesized by using NCA method from L-leucirte and L-phenylalanine. The carboxy group of L-Leu-L-Phe protected by methyl ester was coupled with N-formyl-Met by DCC/HOBT coupling reagents and then methyl group was removed by 0.5 mol·L^-1 NaOH to produce fMLP in the end. Results and conclusion Under these conditions, the total yield of fMLP was 44.0 %. The structure of fMLP was confirmed by IR, IH-NMR, and MS spectra.
