摘要 目的:研究脂质体介导gp130反义寡核苷酸对人雄激素非依赖性前列腺癌(AIPC)细胞PC-3体外增殖活性的影响。方法:采用细胞计数及MTT比色法评价反义寡核苷酸对PC-3细胞体外增殖的影响,应用荧光免疫细胞化学法检测gp130基因的蛋白表达情况,流式细胞计数评价其对细胞周期的影响。结果:脂质体介导gp130反义寡核苷酸组与正义寡核苷酸组及对照组比较,PC-3细胞增殖活性受到明显抑制。转染48h后,gp130蛋白表达完全受到抑制,细胞周期明显阻滞于G0/G1期,细胞增殖抑制率为53.7%,细胞凋亡率高达51.28%。结论:脂质体介导gp130反义寡核苷酸可抑制人AIPC细胞PC-3体外增殖活性。应用反义技术为AIPC的基因治疗提供了新的思路和探索。 Objective: To study the effects of liposome conjugated with gp130 antisense oligonucleotides on proliferation of Androgen independent prostate cancer(AIPC) cell line PC-3. Methods: The trial for the inhibition of cell growth in vitro was conducted to study the effect of liposome conjugated with gp130 antisense oligonucleotides on PC-3 cell(cells count and MTT colorimetry). Then, fluorescence immunocytochemistry technique was performed to detect the protein expression of gp130 gene. Flow cytometric analysis was performed to assay the distribution of cell cycle. Results: The proliferation of PC-3 cell was effectively inhibited by antisense oligonucleotides conjugated with liposome, while the contrast and sense oligonucleotides showed no such effect. The protein expression of gp130 gene was inhibited completely after 48 hours of transfection. Cell cycle was arrested at G0/G1 phase. The inhibited rate of cell proliferation was 53.7% and the the apoptosis rate was as high as 51.28%. Conclusions: The growth activities of AIPC cell line PC-3 and expression of gp130 gene in vitro could be inhibited by gp130 Antisense oligonucleotides conjugated with liposome. It might be an potential gene therapy strategy for AIPC by antisense technique.
