小檗碱对人鼻咽癌CNE-2细胞端粒酶活性的影响 被引量:3

摘要 目的:观察小檗碱(Berberine)对人鼻咽癌CNE-2细胞端粒酶活性的影响。方法:不同浓度的小檗碱处理体外培养的人鼻咽癌CNE-2细胞72 h,聚合酶链反应-端粒重复扩增(PCR-TRAP)法测定端粒酶活性,噻唑蓝(MTT)法测定细胞增殖抑制。结果:小檗碱可抑制CNE-2细胞端粒酶活性,随小檗碱浓度增大,端粒酶活性抑制增强。MTT结果显示小檗碱抑制CNE-2细胞增殖且抑制有浓度依赖性。结论:端粒酶活性的抑制可能是小檗碱发挥抗癌活性的作用机制之一,小蘖碱作为鼻咽癌的辅助治疗可能提高疗效。 Objective: to study the effect of berberine on the tetomerase Activity of human Nasopharyngeal Carcinoma (NPC) CNE-2, Methods:To deal with the human Nasopharyngeat Carcinoma (NPC) CNE-2 which were cultivated outside from the body for 72 hours by different consistence berberine solution in advance , then mensurate the Activity of the telomerase by PCR-TRAP ,and men.surate the inhibition of the cell multiplication by MTT . Results: Berberine can inhibit the telomerase Activity of human CNE-2, enlarging with the berberine density,the inhibition of the telomerase Activity will be built up. MTT showed that berberine can inhibit the multiplication of CNE-2 celt and there is a concentration dependence as a result. Conclusion : To inhibit the Activity of the telomerase maybe is one of the anticancer mechanism of berberine, and it can be one of the adjuvant therapy treating Nasophyt,geal Carcinoma (NPC) maybe it can improve the cure result.
