摘要
目的建立手性分离方法并检查佐米曲普坦光学纯度。方法通过手性拆分剂种类及浓度、缓冲液pH及浓度、温度及电压的优化,选择最佳的手性分离条件。结果最佳分离条件缓冲液为200 mmol/L磷酸盐缓冲液(用磷酸调pH值为2.3,内含21 mmol/L三甲基-β-环糊精);检测波长为214 nm;电压为12 kV;温度为22℃,基线分离了佐米曲普坦及其对映体。结论建立的毛细管电泳法可用于佐米曲普坦的手性分离,为产品的质量控制提供了可靠的分析方法。
Objective To establish method for chiral separation and inspect the purity of Zolmitriptan. Methods By optimizing factors which affect the chiral separation, the kinds and concentration of cyclodextrin, the pH value and concentration of buffer, the voltage and temperature, the optimum conditions for chiral separation were selected. Results Using phosphoric buffer containing trimethyl-β-cyclodextrin, the base-line enantiomers separation of Zolmitriptan was achieved and optical purity was determined. The best conditions were 200 mmol/ L phosphoric buffer, 214 nm detective wavelength and 12 kV voltage. Conclusion The method newly established may be used in the new drug development process for quality control.
