高压氧及低压缺氧预适应对家兔实验性大脑中动脉闭塞后梗塞灶体积的影响 被引量:1

Effects of hyperbaric oxygen and hypobaric hypoxia preconditioning on infarct volume after permanent middle cerebral artery occlusion in rabbits

摘要 目的:观察高压氧预适应(HBOP)及低压缺氧预适应(HHP)对实验性家兔大脑中动脉闭塞(MCAO)后梗塞灶体积的影响,探讨HBOP及HHP的神经保护作用。方法:雄性家兔72只随机分为单纯MCAO组、HBOP+MCAO组,HHP+MCAO组,采用改良Bederson法建立家兔MCAO模型,分别于MCAO后1d、3d、10d及20d观察家兔行为学、病理学及脑梗塞灶体积。结果:行为学评分,单纯MCAO组第1天为3.91±0.14,第3d为3.42±0.47,第10d为2.03±0.56,第20d为1.97±0.35;HBOP+MCAO组和HHP+MCAO组的行为学评分则明显改善(P<0.05),HBOP+MCAO组分别为3.58±0.27(1d)、2.85±0.69(3d)、1.63±0.81(10d)和1.55±1.12(20d),HHP+MCAO组分别为3.47±0.46(1d)、2.78±0.7(3d)、1.5±1.02(10d)和1.41±0.82(20d)。HBOP+MCAO组和HHP+MCAO组的脑梗塞灶体积分别于MCAO后3d、10d和20d也小于单纯MCAO组(P<0.05),单纯MCAO组则为(375.6±15.6)mm3(3d)、(257.5±9.1)mm3(10d)和(231.5±13.9)mm3(20d),而HBOP+MCAO组分别为(332.0±17.6)mm3(3d)、(202.2±12.2)mm3(10d)和(195.3±14.3)mm3(20d),HHP+MCAO组分别为(324.2±15.4)mm3(3d)、(208.1±12.1)mm3(10d)和(201.9±9.9)mm3(20d),但HBOP+MCAO组和HHP+MCAO组之间无明显差异。结论:HBOP和HHP能明显改善实验性MCAO家兔行为学,缩小梗塞灶体积,有一定的神经保护作用。 Objectives. To observe the effects of hyperbaric oxygen preconditioning (HBOP) and hypobarie hypoxia preconditioning (Hi lP) on infarct volume after permanent middle cerebral artery occlusion (MCAO) in rabbits and investigate the neuroprotective effects of HBOP and HHP. Methods: 72 male rabbits were randomly divided into three groups; simple MCAO group (n= 24), HBOP+MCAO group (n=24) and HHP+MCAO group (n= 24). The improved Bederson method was used to establish rabbit permanent MCAO model. The neurological deficits, pathological changes and infarct volumes were detected at 1, 3, 10, 20 d after permanent MCAO. Results;Compared with simple MCAO group, the scores of neurological deficits decreased significantly in HBOP group and HHP group at 1.3.10.20 d after permanent MCAO(P〈0. 05). In the simple MCAO group, the scores were 3.91±0. 14 (ld), 3. 42±0. 47 (3 d), 2. 03±0. 56 (10 d) and 1.97±0. 35 (20 d), but in the HBOP and HHP groups the scores were 3. 58±0. 27/3. 47±0. 46 (1 d), 2. 85± 0. 69/2. 78±0. 7(3 d), 1.63±0. 81/1.5.9±1.02 (10 d ) and 1.55±1.12/1.41±0. 82 (20 d), respectively. The infarct volumes were (332. 0±17. 6)mm^3/(324.2±15. 4)mm^3(3 d),(202. 2±12. 2)mm^3/(208. 1±12. 1)mm^3 (10 d) and(195. 3±14.3)mm^3/(201.9±9. 9) mm^3 (20 d) in HBOP group and HHP group at 1 d,3 d, 10 d, 20 d after permanent MCAO, and they were smaller significantly(P%0. 05)than simple MCAO group whose volumes were 375.6±15. 6 mm^3 (3 d), 257.5 ±9. 1 mm^3 (10 d) and 231.5±13. 9 mm^3 (20 d). Conclusions; HBOP and HHP can prevent neurological deficits and reduce infarct volume after permanent MCAO in rabbits.
