生长激素对人体衰老影响的初步研究

A preliminary study on the influence of growth hormone on the aging process

摘要 目的探讨生长激素对人体衰老的影响。方法12名自愿参加试验研究的健康老年人皮下注射人类生长激素(r-humangrowthhormone,r-hGH),剂量为0.08U/kg,每周3次,共用6个月。结果胰岛素样生长因子(insulin-likegrowthfactor-1,IGF-1)水平注射前均值为762±178U/L,注射3个月后升高为1062±127U/L,差异有显著性(P<0.01)。但注射6个月后,下降为697±124U/L,与3个月相比,差异亦有显著性(P<0.01)。IGF-1下降原因不明。采用双能X线吸收仪(dualenergyX-rayabsorptiometry,DEXA)测定脂肪和精肉,试验前后分别为14559±4182g及13709±3325g和49683±280及50317±2935g,但差异无显著性;骨密度在试验前后变化不显著。结论所有受试者试后均保持健康,除2例6个月血糖稍升高(分别从5.08上升至7.89mmol/L和5.67上升至8.24mmol/L)外,无明显副作用,对应用r-hGH延迟生理衰老的利弊尚需做更多病例观察。 Objective This protocol was designed to investigate the influence of growth hormone (GH) on the aging process. Methods Twelve healthy old volunteers were given GH 0 08U/kg subcutaneously 3 times per week for 6 months. The value of insulin like growth factor 1(IGF 1) was determined with radioimmunoassay. The lean body mass and body fat were measured with dual energy X ray absorptiometry (DEXA). Results The mean value of IGF 1 was 762±178U/L and 1062±127U/L prior to and 3 months after the treatment respectively. There was significant difference between the two values ( P <0 01). However, at the end of 6 months the value declined to 697±124 U/L ; the cause is unknown. There is a tendency of increase in lean body mass and decrease in body fat( P >0 05) as shown by DEXA. There is generally no significant change in bone density but with a little increase in L2 4. Conclusions These results suggest that GH play a role in increasing the lean body mass and decreasing body fat.
