摘要 目的观察已分化神经干细胞植入成年鼠损伤脊髓后的变化及检查修复脊髓损伤的效果。方法以胚胎脊髓提取液(FE)诱导细胞/PGA支架复合物分化后,植入成年鼠损伤脊髓,观察分化细胞的存活和发育情况并评估动物的运动功能。结果FE体外可诱导神经干细胞分化成神经元、星形和少突胶质细胞。植入脊髓后,分化细胞/PGA支架复合物(dNPC)内的细胞成分可以存活并继续成熟,动物BBB评分明显优于对照组。结论dNPC含有已经分化的细胞,移植后各种细胞成分可以存活并继续成熟,对成年鼠损伤脊髓的结构重建和功能恢复都有一定的作用。 Objective To observe the developing of differentiated neural stem cells after grafted into the injuried spinal cord,and to detect the repairing effect. Methods NSCs were implanted into polyglycolic acid (PGA) scaffold,subsequently provid fetal spinal cord extracts (FE) to induce cell differentiation,then it was grafted into the injured spinal cord. Animals were followed for signs of functional recovery by the BBB score,and the survival,growth and mature of these differentiated cells were examinated. Results FE could induce cell differentiation into neurons,astrocytes,and oligodendrocyte. After differentiated neural stem cells/PGA complex (dNPC) implanted into the injured spinal cord,in which cello-reconstituent could survive and kept developing,the BBB score always better than control groups. Conclusion In dNPC the cells have been differentiated,the neurons,astrocyte and oligodendrocyte could survive and became mature,dNPC transplantation might benefit to the animals's behavioral improvement and anatomic reconstruction to certain degrees.
