摘要
目的:通过检测育龄妇女正常月经周期中不同时期的子宫内膜组织、早孕蜕膜和绒毛组织中骨桥蛋白(osteopontin,OPN)的表达,探讨OPN在子宫内膜表达的变化规律及其在胚胎着床过程中的作用。方法:采用免疫组化和逆转录聚合酶链反应等技术,检测正常月经周期的子宫内膜组织、早孕蜕膜、绒毛中骨桥蛋白的表达。结果:OPN在月经周期的各期子宫内膜均有表达,以分泌中期子宫内膜组织的表达最强,且分泌中、晚期的表达量明显高于增生期和分泌早期(P<0.05)。早孕蜕膜和绒毛组织中OPN均呈强表达,明显高于非孕各期子宫内膜组织(P<0.05)。结论:OPN mRNA及其蛋白在分泌中期(种植窗期)子宫内膜的表达明显升高,推测OPN可能与种植窗的开放密切相关。OPNmRNA及其蛋白在早孕蜕膜和绒毛组织中的表达水平进一步升高,提示OPN在胚胎着床后也有重要作用。
Objective :To investigate the expression of osteopontin in reproductive women′s endometrium throughout the menstrual cycle,and early pregnant endometrium and cho- rionic tissue, and discuss the regularity of osteopontin expression in human endometrium and its role during embryo implantation. Methods: The expression of osteopontin in human endometrial specimen and early pregnant endometrium and chorionic tissues was detected by semi-quantita- tive RT-PCR and immuno-histochemistry. Results: Osteopontin mRNA and protein were ex- pressed in human endometrium throughout the menstrual cycle. The expression levels of os- teopontin mRNA and protein were all significantly higher in human mid and late secretory endo- metrium compared with proliferative endomtrium and early secretory endometrium ( P 〈 0.05 ). Osteopontin was also expressed in early pregnant endometrrium and chorionic tissues. Moreover, the expressive quantity of osteopontin mRNA and protein were significantly higher in these tis- sue than that in nonpregnant endometrium (P 〈 0.05 ). Conclusions: The expression of os- teopontin mRNA and protein are increased in human mid secretory endometrium (around the time of “implantation window” ), indicating that osteopontin may participate in opening “im- plantation window”. Osteopontin mRNA and protein are increased further in early pregnant en- dometrrium and chorionic tissues, presumed that osteopontin may play a critical role during the time of after embryo implantation.
