摘要
采用高聚物型苯乙烯-二乙烯基苯(PS—DVB)麦科菲反相高效液相色谱柱(MKF—RP—MH)分离胸腺素α1,优化色谱条件为:流动相A:0.01mol/L磷酸盐缓冲液(pH7.0或8.0),流动相B:乙腈,梯度洗脱条件0~20min,0%~10%乙腈,20~40min,10%~30%乙腈;温度:30℃;流速:0.8mL/min;检测波长:214nm。在优化条件下分离了胸腺素α1标准品和样品。样品浓度在0.05~4g/L时,线性方程为C=0.0099A-0.0798(r=0.9994),胸腺素α1最大载样量为120μg(6g/L)。
A reversed phase high performance liquid chromatographic (RP-HPLC)method with polystyrene- divinylbenzene (PS-DVB) resin (MKF-RP-MH) was developed for the determination of thymosin α1. In the method,thymosin α1 was chromatographically analyzed with a mobile phase A: 0.01mol/L phosphate buffer solution ( pH = 7.0 or 8.0) and B : acetonitrile, linear gradient elution condition : 0 - 20 rain, 0% - 10% acetonitrile, 20 - 40 min, 10% - 30% acetonitrile; column temperature was set at 30℃ ; flow rate was 0.8 mL/min; The detection wavelength was 214 nm. The standard sample and two kinds of thymosin α1 samples were analysised under the optimum chromatographic condition. The calibration curve was in good linearity within the range of 0.05 - 4.0 g/L with the regression equation of C = 0. 0099A - 0. 0798 ( r = 0. 9994). The maximum amount of thymosin α1 on MKF-RP-MH column was 120 μg(6 g/L).
