氯胺酮预先给药对脂多糖诱导大鼠肺泡巨噬细胞超氧阴离子生成和细胞内钙离子浓度的影响

摘要 脂多糖(LPS)是革兰氏阴性菌外膜的组成成分,侵入机体后,在LPS结合蛋白介导下,与巨噬细胞膜上的CD14结合,通过NF-κB信号转导途径促进细胞因子释放和超氧阴离子(O2^-·)生成,诱发炎性反应。巨噬细胞是炎性反应的主要效应细胞,在全身炎性反应综合征时通过释放O2^-·和肿瘤坏死因子.α等细胞因子诱发炎性反应。氯胺酮是非竞争性N-甲基-D-天门冬氨酸(NMDA)受体拮抗剂,能够与NMDA受体离子通道结合,阻断Ca^2+通道。氯胺酮可抑制巨噬细胞释放炎性因子,其作用机理是否与阻断Ca^2+通道从而抑制O2^-·生成,进而发挥抗炎作用有关尚待进一步探讨。本研究拟探讨氯胺酮对LPS诱导大鼠肺泡巨噬细胞O2^-·生成和细胞内Ca^2+浓度的影响。
