摘要
目的研究淋巴瘤和反应性增生淋巴结的基因表达差异。方法将淋巴瘤淋巴结标本和反应性增生淋巴结标本的mRNA与表达谱芯片杂交,获得表达差异基因。用生物信息学方法对新基因C14ORF44进行初步分析。结果表达谱芯片共找出了152条差异表达基因。用生物信息学方法分析了C14ORF44的基因结构、染色体定位、编码蛋白质的理化性质、亚细胞定位等信息。对C14ORF44蛋白的多物种相似蛋白进行了系统进化分析,基本明确了该基因编码一个在胚胎和肿瘤细胞中高表达、进化中保守的核蛋白。结论生物信息学分析表明,C14ORF44是一个有研究价值的癌相关蛋白。这个蛋白可能参与正常的胚胎发育,但也与肿瘤形成有关。
Objective To explore the difference of expressed genes of lymphoma and reactive lymph node. Methods Laser capture microdissection was combined with genechip analysis. An initial bioinformatic analysis was performed on a novel gene, C140RF44, which is highly expressed in lymph node of lymphoma, mRNA of the tissue from lymphoma and reactive lymph node was labelled with biotin respectively and hybridized with expression profile microarray, and the expressed genes were obtained. Initial bioinformatic analysis was performed on a novel gene named C140RF44, which has no functional researches at present. Results With expression profile microarray, many differential expressed genes were uncovered. Bioinformatic analysis has been performed on C140RF44. Its gene structure, genomic localization, the physical and chemical characteristics of the putative protein, subcellular localization, functional domain and so on are predicted, and the systematic evolution analysis was performed. Efficient bioinformatic analysis has fundamentally identified that C140RF44 is a nucleus protein, which is highly conservative in many species. Conclusion Bioinformatic analysis of C140RF44 showed that it was a valuable potential tumor associated protein. The protein may be involved in the embryogenesis and tumor formation.
