顽固性癫痫痫性皮层组织差异表达基因的研究 被引量:3

Studies of differential genes expression of epileptic cortexes: fluorescent differential display PCR analyses

摘要 目的应用荧光mRNA差异显示PCR技术对比研究痫性皮层及非痫性皮层组织基因的差异表达,探讨该差异与癫痫发病机制相关的分子生物学基础。方法取8例痫性皮层组织(其中3例隐源性癫痫痫性皮层、5例继发性癫痫痫性皮层)和5例非痫性脑皮层组织的冰冻标本,应用荧光mRNA差异显示。PCR技术比较两种组织的基因表达差异。结果痫性皮层与非痫性皮层组织中存在明显的基因表达差异。结论(1)荧光mRNA差异显示PCR技术是研究痫性皮层差异表达基因的一种行之有效的方法;(2)痫性皮层和非痫性脑皮层组织之间存在明显的mRNA表达差异; (3)差异显示片段Z4和Z8在非痫性皮层组织中高表达,痫性皮层组织中低表达,基因片段所代表产物的分别是Cu/Zn超氧化物歧化酶和蛋白磷酸酶,均位于人类第5号染色体长臂上,可能是癫痫的相关基因。 Objective To investigate differentially expressed genes between epileptogenic cortex in patients with intractable epilepsy and normal cortex with fluorescent differential display reverse transcription polymerase chain reaction ( DD-PCR/DDRT-PCR ), so as to explore the correlation between differentially expressed genes and epileptic pathogenesis on molecular biology. Methods This study included 8 epileptogenic cortexes and 5 normal cortexes. The total mRNA from frozen specimens was extracted and the differentially expressed genes were compared by fluorescent DDRT-PCR. Results Significant differences of gene expression were detected between the epileptogenous cortex and normal cortex. Conclusions (1) Fluorescent DDRT-PCR is a useful method to detect and characterize altered gene expression in epileptogenic cortexes and normal brain tissue. (2) There are significant differences of gene expression between the epileptogenic cortex and normal cortex. (3) Differentially displayed band Z4 and Z8 which sequences located in the human chromosome 5 are expressed lower in epileptogenic cortexes. It is supposed that the superoxide dismutase and protein phosphatase l H may play a role in the development epilepsy depending on the regulation of gene expression.
