兔蛛网膜下腔出血后NF-κB在基底动脉的表达 被引量:8

The expression of NF-κB in the basilar artery of rabbits after SAH

摘要 目的探讨兔蛛网膜下腔出血(SAH)后核转录因子-κB(NF-κB)的表达与脑血管痉挛(CVS)的关系。方法新西兰纯种大白兔160只,枕大池二次注血,制作兔SAH后CVS的模型。分离基底动脉,应用形态学观察、免疫组化、原位杂交等方法,观察兔基底动脉管径变化、血管壁上NF-κB的表达及其与CVS的关系。结果在SAH后第7天,DSA证实基底动脉痉挛明显;痉挛血管壁上NF-κB的表达在SAH后第5天出现强烈表达,两者的表达有时项上的差别。抑制NF-κB的表达可以有效缓解CVS的发生。结论NF-κB的表达与CVS的发生有一定的相关性,在CVS的发生和发展过程中起了重要的作用。 Objective To discuss the existence of NF-κB in the occurrence and progress of CVS in the basilar artery of rabbits and the relations of them. Methods 160 New Zealand pure rabbit were afforded. Models of SAH and CVS were successfully made via double blood injection into major cistern and basilar artery were separated. Morphological methods and immuno- histochemistry were employed to observe the variance of basilar artery and dynamic expression of NF-κB in the wall of the basilar artery. Results Stenosis of basilar artery got to the top in 7d which was accordance with morphological observation and could afford foundation for the observation of patho-physiological changes of CVS. The expression of NF-κB alter accompanied with the stenosis of spasmodic artery. The expression of NF-κB was significiantly high in 5d after SAH and had difference in sequence and had close correlation with the happening and progress of CVS. The successful inhibition of NF-κB could relax the occur of CVS. Conclusion Inflammation of spasmodic artery controlled by NF-κB do exist in the happening and progress of cerebral vasospasm. These relations played an important functions in the happening and progress of CVS.
