摘要 目的观察白细胞介素(IL)-4、IL-13在恶唑酮诱导的小鼠实验性结肠炎中的变化,并探讨其在溃疡性结肠炎(UC)中的作用机制。方法健康昆明小鼠24只随机均分为正常组和模型组(以恶唑酮灌肠造模)。造模3d后全部处死,提取外周血单个核细胞(PBMC)、脾脏单个核细胞(SMC)和肠黏膜固有层单个核细胞(LPMC)。进行细胞培养后,采用荧光定量聚合酶链反应检测IL-4、IL-13的表达量。结果培养24h后,模型组SMC、LPMC和PBMC中IL-4的表达量均显著高于正常组(P值均<0.01),SMC和PBMC中IL-13的表达量均显著高于正常组(P值分别<0.05、0.01)。结论IL-4和IL-1 3在UC的发病机制中起重要作用。 Objective To investigate the changes of interleukin(IL)-4 and IL-13 expression in oxazoloneinduced colitis in mice and to explore the role of IL-4 and IL-13 in the development of ulcerative colitis(UC). Methods Twenty-four healthy Kunming mice were randomized into two groups(n = 12) : normal control group and model group. The mice in model group were induced by oxazolone enema. All mice were sacrificed 3 days after enema, and their peripheral blood mononuclear cells(PBMC), spleen mononuclear cells(SMC) and lamina propria mononuclear cells(LPMC) were harvested from freshly tissues. Expression of IL-4 and IL-13 in SMC, LPMC and PBMC was determined by fluorescence quantitative polymerase chain reaction(PCR) after culture. Results IL-4 epxression in SMC, LPMC and PBMC were significantly higher in model group than in normal control group 24 h after culture(all P 〈 0.01). IL-13 expression in SMC(P 〈 0.05) and PBMC(P 〈 0.01) was significantly higher in the model group than in normal group. Conclusion The results indicate that IL-4 and IL-13 are deeply involved in the mechanisms of UC. (Shanghai Med J, 2007, 30:774-776)
