干扰素γ/Fas介导的HaCaT细胞凋亡及8种人参皂甙的作用 被引量:4

摘要 目的体外诱导角质形成细胞凋亡并研究人参皂甙对细胞凋亡的影响。方法将IFN-γ,Anti Fas Ab作用于体外培养角质形成细胞HaCaT诱导细胞凋亡,并检测人参皂甙对细胞凋亡的影响。结果50 ng/mL IFN-γ和25 ng/mL Anti Fas Ab共同作用可以诱导21.5%角质形成细胞HaCaT凋亡。8种人参皂甙中,Mx,CK和Re可逆转Anti Fas Ab诱导的HaCaT细胞凋亡,逆转率分别为18.4%,14.5%和9.6%。结论IFN-γ和Anti Fas Ab共同作用可诱导HaCaT细胞凋亡。人参皂甙Mx,CK和Re对Anti Fas Ab诱导的HaCaT细胞凋亡有一定的抑制作用。 Induced HaCaT cells apoptosis by INF-yand Anti-Fas monoclonal antibody (Anti Fas Ab) and study potential therapeutic effect of ginsenosides. Methods The keratinocyte cell line, HaCaT,was exposed to IFN-γ and AntiFas Ab, and observed effects of ginsenosides on apoptosis . Results Flow cytometry demonstrated that IFN -γ and AntiFas Ab induced 2 1. 5 % HaCaT apoptosis. Ginsenosides Mx, CK and Re, among eight ginsenosides screened, inhibiting KC apoptosis 1 8.4 % , 14.5% ,9.6% respectively. Conclusion Combination of IFN-γ and Anti Fas Ab induces the apoptosis of HaCaT cells. Ginsenosides Mx,CK,and Re inhibit the apoptosis of HaCaT cells.
