摘要
目的了解徐州地区小肠结肠炎耶尔森菌的分布及病原学特征。方法常规方法在腹泻患者粪便、家禽家畜粪便、苍蝇、肉食品中分离小肠结肠炎耶尔森菌,用血清学、生物学方法分型及抗菌耐药性分型。并通过PCR技术进行毒力基因检测,同时对检出的国内主要流行血清型O:3和O:9菌株进行脉冲场凝胶电泳分析。结果2014份标本中共检出32株(分为17个血清型)小肠结肠炎耶尔森菌,总检出率为1.59%,其中O:3和O:9血清型的检出率分别为9.4%,且耐药模式相同。32株小肠结肠炎耶尔森菌分别携带ail、ystA、yadA、virF和ail、ystA毒力基因的各占9.4%,只携带ystB基因的占21.8%,不携带以上任何基因的占59.4%。32株小肠结肠炎耶尔森菌分为3个生物型别,其中1A型为78.1%,3型为18.8%,2型为3.1%。本地区分离的2株O:3血清型小肠结肠炎耶尔森菌的脉冲场凝胶电泳带型相同,是我国O:3血清型小肠结肠炎耶尔森菌的主要克隆系。结论徐州地区存在致病性小肠结肠炎耶尔森菌O:3和O:9血清型及由此引起的腹泻疾病,应引起高度重视。
Objective To investigate the etiology and distribution of Yersinia enterocolitica in Xuzhou area. Methods Yersinia strains were isolated from the feces of patients with diarrhea, live stock and poultry, as well as from flies and meat products using conventional methods. The isolates were tested for serologic, biologic types and drug resistance. Polymerase chain reaction(PCR) technique was used to detect virulence gene. At the same time, the detected O : 3 and O : 9 strains, which were the main epidemic strains in China, were analyzed using pulsed-field gel electrophoresis (PFGE). Results Thirty-two strains(17 serotypes) of Yersinia enterolitica were detected from 2 014 samples. The total isolation rate was 1.59%, among which the isolation rate of O : 3 and O : 9 sero types that have the same mechanism of drug resistance was 9.4 %. Among the 32 strains, the strains carrying ail, ystA, yadA, virF virulence genes or the strains carrying ail, ystA virulence genes both counted for 9.4%. Only ystB gene positive strains were 21.8%. The strains with none of the above genes were 59.4%. The 32 strains of Yersinia enterolitica were divided into 3 biological types, among which type 1A was 78. 1%, type 3 was 18.8%, and type 2 was 3. 1%. The fragments of PFGE were the same between the 2 isolates of O : 3 serological type of Yersinia enterolitica isolated in this area, which was the main clone of O : 3 serological type in China. Conclusion There are pathogenic strains of O : 3 and O : 9 serological types of Yersinia enterolitica and related diarrhea diseases in Xuzhou area, which need much attention.
