利用热水处理结合心叶愈伤组织培养脱除甘蔗宿根矮化病菌研究 被引量:14

摘要 甘蔗宿根矮化病(Ratoon Stunting Disease,RSD)是我国蔗区普遍发生且造成严重经济损失的一种甘蔗病害,研究其防治措施非常重要。本文应用50℃2h热水处理感病种茎,以其长成植株的心叶为外植体进行愈伤组织培养,应用斑点酶标记免疫试验(Dot blot enzyme immunoassays,DB-EIA)对愈伤组织培养瓶苗、假植苗、再生植株蔗茎进行RSD病菌检测,以检验该种方法的脱菌效果。结果表明:瓶苗、假植苗、再生植株蔗茎均未能检测出RSD病菌,该法可有效的去除甘蔗RSD病菌,且经田间观察再生植株生长健壮、未发现变异植株。 Sugarcane Ratoon Stunting Disease (RSD) is known to be an economically important disease in China. It is necessary to study the method for controlling this disease. Using RSD-infected seedcane as material, the method of eliminating causal bacterium-of RSD, Leifsonia xyli subsp. Xyli (Lxx) was studied by combining hot water treatment (50℃ for 2 hours) with callus culture using young leaves as explants. Dot blot enzyme immunoassays (DB-EIA) was employed to confirm the eliminating results of RSD. The results from DB-EIA assay of the regenerated plantlets in medium culture stage, in nursery stage and the mature cane in the field showed that this method was very effective for getting rid of the disease. The field observation of the crop developed from the disease-free seedlings indicated that the cane plants replants grew well without any variation.
