摘要 【目的】了解葡萄糖腹膜透析液(PDS)对体外培养人腹膜间皮细胞凋亡的影响。【方法】通过向培养液中添加1.5%葡萄糖腹膜透析液,观察细胞caspase-3活性变化,应用Annexin V/PI双染色流式细胞仪方法以及TUNEL方法观察腹膜间皮细胞凋亡的情况。【结果】PDS组细胞caspase-3活性较对照组明显上升。Annexin V-FITC/PI双标记结果显示PDS组细胞凋亡率为(46.8±14.2)%,显著高于对照组的(9.04±4.86)%(P<0.05)。TUNEL结果显示PDS组细胞凋亡明显高于对照组。【结论】葡萄糖腹膜透析液可以明显诱导腹膜间皮细胞细胞凋亡的发生。 [Objective]To investigate the apoptosis of cultured human peritoneal mesothelial cells induced by peritoneal dialysis solution (PDS). [Methods]Using RPM1640 culture medium as control , commercial PDS containing 1.5% glucose were added into the culture medium to investigate the apoptosis of cultured human peritoneal mesothelial cells induced by peritoneal dialysis solution . The activity of caspase-3, the terminal deoxynucleotidyl transferase (TdT) mediated dUTP nick end labeling (TUNEL) and flow cytometry analysis were used to describe the apoptosis rate of each group in this study. [Results]The activity of caspase-3 in control group was defined as 1, the relative activity of caspase-3 in PDS group was elevated to 3. 26±0.91. The apoptosis rate detected by PI/Annexin V in PDS group was 46.8±14.2% which was significant higher than that in the control group. Higher apoptosis rate detected by the method of TUNNEL was also found in PDS group than that in the control group. [Conclusion]Marked apoptosis can be induced by : Commercial PDS in cultured human peritoneal mesothelial cells in vitro.
