摘要
肿瘤抑素(tumstatin)是继内皮抑素(endostatin)和血管生成抑素(angiostatin)发现以来,来源于人血管基膜Ⅳ型胶原蛋白的极具潜力的肿瘤抑制因子。它强大的抗肿瘤新生血管生成、诱导肿瘤细胞和内皮细胞凋亡等生物活性,使其成为近年来的研究热点。本文详细介绍了肿瘤抑素活性肽段及其衍生物的研究进展。对Tum-5、T7肽和19肽以及这些活性肽段连接NGR导向肽或人源性IgG3铰链区等肽段后形成的衍生物作了较详细的介绍。肿瘤抑素的活性肽段及其衍生物,以高选择性、高活性、低毒性等优点正成为抗肿瘤药物研究的新希望。
After the development of angiostatin and endostatin,tumstatin,which is derived from human vascular basement membrane component-type Ⅳ collagen,came out as a tumor inhibitory factor with great potential.Its bioactivities,such as antitumor angiogenic activity,apoptosis induction of tumor cells and endothelial cells,have made it to be a hot research spot.In this article,the advance in research on tumstatin active peptides and their derivatives are introduced and the bioactivities of Tum-5,T7 and 19 peptides and the derivants of these active peptides linking with NGR sequence or human IgG3 hinge region are summarized in detail.Tumstatin active peptides and their derivants have been being promising candidates for the treatment of cancer,showing their features of high selectivity and activity as well as low toxicity.
