CpG-ODN在T细胞活化和抑制中的作用

The roles of CpG ODN on T lymphocyte activation and suppression

摘要 CpG-ODN 是人工合成的含有 CpG 基序的 ODNs,能够模拟细菌 DNA 的免疫刺激活性,通过细胞内定位的 Toll 样受体9活化抗原递呈细胞(APC),如树突状细胞(DC)和 B 细胞,分泌大量 Th1型细胞因子诱导产生 Th1型免疫应答。CpG-ODN 不仅增强 T 淋巴细胞的活化,在特定条件下还可以抑制 T 淋巴细胞功能。在抗感染、抗肿瘤疾病中,CpG-ODN 作为新型的免疫佐剂得到了广泛的应用,但其安全性问题应引起足够的重视。 Synthetic oligodeoxynucleotides (ODNs) containing CpG motifs mimic the activity of bacterial DNA. It is generally accepted that CpG oligodeoxynucleotides ( CpG-ODNs) are recognized by Toll-like receptor 9, which is localized in the cytoplasmic follicle of antigen presenting cells, such as dendritic cells and B cells, subsequently result in those cells to proliferate, mature, and secrete a variety of cytokines, chemo- kines predominantly driving Thl polarized T-cell immune responses. CpG-ODNs not only enhance the activation and maintenance of T lymphocyte, but also play an important role on the suppression of T lymphocyte. Even though CpG-ODNs have been extensively applied as potent immunological adjuvants against infections and cancers, more attentions should be paid to the safety of CpG-ODNs.
