摘要
目的:观察芍药苷对辐射致血虚小鼠骨髓细胞蛋白质表达的作用,探讨芍药苷生血作用的分子机理。方法:采用3.5Gy^(60)Coγ射线全身1次性照射制备小鼠血虚模型;利用蛋白质组学技术,分离、分析、鉴定差异表达蛋白质。结果:芍药苷能部分逆转辐射致血虚证小鼠骨髓细胞蛋白质表达的改变,使5种蛋白质表达回升,7种回落。结论:芍药苷作用于骨髓细胞多个蛋白质靶点。芍药苷调节骨髓蛋白质表达的作用可能是其促进骨髓造血细胞生长、增殖和分化,发挥生血作用的重要机理之一。
To observe effect of paeoniflorin on protein expression of bone marrow cell of blood deficiency mice induced by radiation and study the molecular mechanism of paeoniflorin in blood enriching function. Methods: The blood deficiency mice were induced by using a single dose of 3.5 Gy 60Co γradiation. Proteomic methods were used to search differential expression proteins. Results: Paeoniflorin can reverse partly the change of protein expression induced by irradiation, which making five kinds of down - regulated proteins raise again and seven kinds of up - regulated proteins fall back of mice bone marrow cell. The proteins raised again include LSP1, proteasome 26S ATPase subunit 4, H- Ras and glyceraldehyde- 3 -phosphate dehydrogenase. The proteins fallen back include HCP, HDAC3, Krox- 6.1A, Grbl4 and an unnamed protein. Conclusion: Paeoniflorin can act on multiple protein targets of bone marrow cells. The effect of paeoniflorin on regulating marrow protein expression could be one of main mechanisms that paeoniflorin can promote hemopoietic cell growth, proliferation and differentiation and then act its blood enriching function.
