垫状卷柏中双黄酮抑制黄嘌呤氧化酶的活性研究 被引量:12

摘要 目的:探寻垫状卷柏中抑制黄嘌呤氧化酶的活性部位。方法:采用黄嘌呤氧化酶体外抑制实验测定各组分的半数抑制浓度IC50。结果:三个双黄酮成分的混合物Fr5对黄嘌呤氧化酶有强抑制作用(IC50=1.1μg/ml),Fr5继续分离得到的两个组分Fr5-1(IC50=2.9μg/mL)和Fr5-2(IC50=24.0μg/mL),其纯度增高但活性不及Fr5,当重新组合这两个组分时,活性仍然不能恢复或增强。结论:垫状卷柏中对黄嘌呤氧化酶抑制作用最强的部分是三个双黄酮类成分的混合物,继续分离活性减弱。 To determine the inhibiting xanthine oxidase active parts of Selaginella pulvinata. Methods : Screen out the active parts by measuring the 50% inhibitory concentration (IC50) of the each fraction against xanthine oxidase in vitro. Results: The biflavonoid fraction Fr5, which contains three bifavonoids, was most active in inhibiting XOD( IC50 = 1.1μg/ml), the Fr5 - 1 (IC50 = 2.9μg/ml) and Fr5 -2 (IC50 = 24.0μg/ml) were subfractions isolated from Fr5, they were more pure but less active than Fr5 ,when mixed the two subfractions again,the antioxidant activity could not be recovered or increased. Conclusion: The most active antioxidant part in Selaginella pulvinata is the mixture of three biflavonoids, a further isolation will bring less activity.
