大黄素抑制增生性瘢痕成纤维细胞胶原合成及细胞周期的研究 被引量:3

摘要 目的检测大黄素对人增生性瘢痕成纤维细胞的抑制作用,探索大黄素防治瘢痕的机制。方法进行人增生性瘢痕成纤维细胞的体外培养,将不同浓度的大黄素作用于细胞,以3H-脯氨酸掺入法检测成纤维细胞胶原合成的情况,并以流式细胞术检测成纤维细胞的细胞周期。结果在不同浓度组大黄素的处理下,人增生性瘢痕成纤维细胞的胶原合成呈现出抑制的趋势;大黄素可以提高细胞G1期比例,而降低S期和G2期比例。结论大黄素对人增生性瘢痕成纤维细胞胶原合成有显著的抑制作用,其机制和细胞周期阻滞在G1期从而抑制细胞增殖有关。 Objective To investigated the inhibitation of to emodin Hypertrophic Scar Fibroblasts,which could be helpful to prevent and therapy the Hypertrophic Scar. Methods The human Hypertrophic Scar fibroblasts was cultured in vitro, different concentration of Emodin (20μg/ml,40μg/ml,60μg/ml) was added in the culture medium. :^3H-proline was used to detective the collagen anabolism,and flow cytometry was used to explore the ceil cycle. Results With the different concentration of emodin treatment, effect of an inhibition ratio on collagen anabolism rised up. The cell cycle was significantly blocked at G1 phase,and appeared dose-dependent maner. Conclusion Emodin could inhibit the collagen anabolism of hypertrophic scar fibroblast cell, which may be caused by Emodin blocking the cell cycle to inhibit the cell proliferation with dose-dependent.
