男女腹部皮下和网膜脂肪中脂联素、肿瘤坏死因子α、过氧化物酶体增殖物激活受体γ_2 mRNA表达的比较

摘要 目的比较男女腹部皮下脂肪组织、网膜脂肪组织中脂联素、肿瘤坏死因子-a(TNF-a)、过氧化物酶体增殖物激活受体γ2(PPARγ2)mRNA表达。方法选择30例择期外科手术患者,男15例,女15例,用RT-PCR法检测腹部皮下脂肪组织和网膜脂肪组织中脂联素、TNF-a、PPARγ2 mRNA表达。结果(1)男性皮下脂肪组织中脂联素mRNA显著低于女性,网膜脂肪组织中TNF-a mRNA显著高于女性(均P〈0.05)。(2)女性皮下脂肪组织中脂联素mRNA表达高于其网膜脂肪组织,男女网膜脂肪组织中TNF-a mRNA均高于各自的皮下脂肪组织(P〈0.05~0.01)。(3)男女间皮下脂肪组织和网膜脂肪组织中PPARγ2 mRNA表达差异无统计学意义(P〉0.05)。结论人腹部皮下脂肪组织和网膜脂肪组织中脂联素、TNF-a mRNA的表达可能存在性别差异,而PPARγ2 mRNA表达的性别差异则不明显。 Objective To investigate the differences of mRNA expressions of adiponectin(APN) ,tumour necrosis factor alphal(TNF-α) and peroxisomal proliferator-activated receptor,/2 (PPARγ2) between male and fe- male in human abdominal subcutaneous and omental adipose tissue. Methods 30 patients to be operated were divided into male group(n = 15) and female group(n = 15). The mRNA expressions of APN ,TNF-α and PPARγ2 in abdominal subcutaneous and omental adipose tissue were measured using RT-PCR method. Results ( 1 ) APN mRNA expression in subcutaneous adipose tissue was lower in male than in female, meanwhile TNF-α mRNA expression in omental adipose tissue was higher in male than in female (all P 〈 0.05). (2) APN mRNA expression in subcutaneous adipose tissue of female was higher than that in omental adipose tissue. No matter in male and in female group, TNF-α mRNA expression was higher in omental adipose tissue than in subcutaneous adipose tissue (P 〈 0, 05 - 0.01 ). (3) No significant differences between male and female groups were found for PPARγ2 mRNA expression in both subcutaneous and omental adipose tissue (P 〉 0.05). Conclusions There might be a signifi- cant difference in mRNA expressions of APN and TNF-α, except for PPARγ2 mRNA expression , between male and female in human abdominal subcutaneous and omental adipose tissue.
