COX-2在大鼠全肝/部分肝移植缺血再灌注损伤中的表达 被引量:3

EXPRESSION OF COX-2 IN ISCHEMIA REPERFUSION INJURY OF WHOLE OR PARTIAL LIVER TRANSPLANTATION IN RAT

摘要 目的探讨COX-2在大鼠全肝/部分肝移植缺血再灌注损伤(IRI)模型中的表达及其意义。方法雄性Wistar大鼠随机分成3组:正常对照组(O);全肝移植组(LT);50%部分肝移植组(PLT)。逆转录-聚合酶链反应(RT-PCR)半定量检测COX-2 mRNA的表达;免疫组织化学显示COX-2阳性细胞分布规律;酶联免疫吸附试验(ELISA)检测血清TNF-α和IL-10水平。结果COX-2的表达为肝移植缺血再灌注损伤机制所必需,IRI损伤较重的部分肝移植组(PLT)伴随有较高水平的COX-2 mRNA表达;且存在其表达量随再灌注后时间点的延长而逐渐下降的规律。结论COX-2参与了肝移植缺血再灌注损伤,是移植物IRI的重要效应分子,是IRI的参与者之一。 Objective To investigate the expression of COX-2 in ischemia reperfusion injury of whole or partial liver trans-plantation in rat. Methods Male Wistar rat were divided randomly to 3 groups : ormal group(O), liver transplantation group( LT), and 50% partial hver transplantation group(PLT). The expression of COX-2 mRNA and protein were detected by RT-PCR and immu-nohistochemistry respectively. And the level of TNF-α and IL-10 were detected by ELISA after operation. Results The expression of COX-2 is necessary to ischemia reperfusion injury after liver transplantation and there is a high level expression of COX-2 mRNA in partial liver transplantation group which ischemia reperfusion injury is heavier than whole liver transplantation group. And the expression of COX-2 mRNA is weak with the time past after reperfusion. Conclusion COX-2 takes part in the ischemia reperfusion injury of hver transplantation and is an important factor in IRI of graft. And COX-2 is one of participator in IRI.
