摘要
[目的]为了研究溶剂萃取法提取茶叶中儿茶素的优化条件。[方法]以乙醇为浸提液,采用香草醛-盐酸法测定茶叶中儿茶素的含量,并通过正交实验确定提取茶叶中儿茶素的最佳工艺条件。[结果]儿茶素溶液的最大吸收波长为505 nm。儿茶素浓度与吸光度间的回归方程为:A=0.017 5X+0.044 7(R2=0.999 7)。正交实验结果表明,儿茶素的最佳提取和纯化条件为:以80%的乙醇为溶剂浸提2次,60℃下浸提35 min,料液比为1∶13。影响儿茶素含量的因素依次为:乙醇浓度>浸提时间>提取温度>料液比。[结论]该研究为儿茶素的应用奠定了基础。
The research aimed to investigate the optimum conditions for extracting catechin in tea by solvent extraction method.[Method]With ethanol as leaching liquor,the method of vanillin-hydrochloric acid was used to determine the content of catechin in tea and the optimum technique conditions for extracting catechin in tea was confirmed by orthogonal test.[Result]The maximum absorption wavelength of catechin solution was 505 nm.The regression equation between catechin concentration and absorbance was:A=0.017 49X+0.044 73(R2=0.999 7).The results of orthogonal test showed that the optimum extracting and purifying conditions for catechin were as follows: extracting with 80% ethyl alcohol as solvent for 2 times,extracting at the temperature of 60 ℃ for 35 min,the ratio of material to liquid of 1∶13.The factors that affected the content of catechin were as follows: ethanol concentration〉extraction time 〉 extraction temperature 〉 the ratio of material to liquid.[Condusion]The research laid the foundation for the application of catechin.
