摘要
目的研究并优化建立鼻咽癌移植瘤动物模型的成熟方法,观察移植瘤的肿瘤生物学特性。方法采用免疫组化(SP)法,对鼻癌细胞株CNE-2对数生长期瘤细胞爬片行EB病毒潜伏膜蛋白2A(LMP-2A)免疫组化检测。对数生长期瘤细胞,分别以5×105/0.2ml,1×106/0.2ml,2×106/0.2ml,4×106/0.2ml及肿瘤组织接种于裸小鼠右侧前肢肩背部皮下,观察移植瘤生物学特性,并取肿瘤行常规病理切片及LMP2A免疫组化检测。结果BALB/c裸小鼠接种CNE-2细胞后,2×106/0.2ml组和4×106/0.2ml组移植瘤成功,3周左右接种部位皮下出现肉眼可见的瘤体肿块,逐渐增大,约4周进入加速期,其致瘤成功率为83.3%;瘤块包埋的裸鼠移植瘤成活率100%,无潜伏期,成功率显著高于另两组(P<0.01)。经病理及免疫组化证实移植瘤成功。病理提示肿瘤长至直径约1.0cm以上时,瘤体中央见坏死区域。结论皮下接种CNE-2细胞成瘤的难易与肿瘤细胞浓度成正相关,2~3×106/0.2ml的肿瘤细胞接种是较为理想的浓度。当肿瘤直径为0.5~0.8cm大小时进行干预研究是理想时机。
Objective To investigate and optimize the method of the establishment of NPC animal model, and to observe biologic characteristics from NPC. Methods The expression of EBV LMP2A in CNE-2 was detected by S-P immunohistochemical methods. BALB/C nude mouse were subcutaneously injected with 5×10^5/0.2ml, 1×10^6/0. 2ml, 2×10^6/0. 2ml, 4×10^6/0. 2ml CNE-2 cells to establish NPC animal models. The tumor characteristics and pathological features were observed. Results NPC animal models were established successfully, which was proved by pathological finding and S-P immunohistochemistry. The tumor tissues of mouse revealed positive expression of EBV-LMP2A, According to pathological finding, necrogenous reaction was observed in the middle of the tumor when the diameter of the tumor was over 1.0cm. Conclusions The concentration of CNE-2 ceils, 2-3 ×10^6/0. 2ml, is the best choice to build NPC animal models, and it suggests that we should intervene the tumor model when the diameter of the tumor is between 0.5cm and 0.8cm.
