摘要 目的:通过观察卡托普利对2型糖尿病大鼠血管内皮功能的影响,探讨卡托普利对糖尿病微血管并发症的防治机制。方法:采用小剂量链脲佐菌素尾静脉注射加高糖高热量饲料喂养的方法建立2型糖尿病大鼠模型,将模型动物随机分为模型组(DM组)、卡托普利组(C组)(50 mg·kg^(-1)·d^(-1)),另设正常对照组(N组)。药物干预治疗12周后。检测各组动物空腹血糖(FBG)、总胆固醇(TC)、三酰甘油(TG)、高密度脂蛋白-胆固醇(HDL-C)、24 h尿微量白蛋白(Alb)、尿α_1微球蛋白(α_(1-m))及各组动物血浆中内皮素(ET-1)、血栓素B_2(TXB_2)、6-酮前列腺环素(6-keto-PGF_(1α))、一氧化氮(NO)的表达水平。结果:与模型组相比,卡托普利组FBG、TC、TG、HDL-C值未见明显改变,而Alb、α_1-m量明显减少,血浆中ET-1、TXB_2的表达水平、TXB_2/6-keto-PGF_(1α)比值显著下降。结论:卡托普利能明显改善血管内皮功能,这可能是其防治糖尿病微血管并发症的机制之一。 Objective : To investigate the effects of captopril on vascular endothelial function in type 2 diabetic rats and explore the protective mechanism of captopril in type 2 diabetic microangiopathy. Method: The routine with type 2 diabetes models were induced by the intravenous injection of a small dose of streptozotocin plus high fat and high caloric laboratory chow. Then modeled diabetic animals were divided into model group, catopril group. Normal rats fed with routine chow were designated as normal group. 12 weeks after treatment , the levels of fasting blood glucose (FBG) , serum total cholesterol (TC) , triglycerides (TG) , high density lipoprotein- cholesterol (HDL-C), 24 hours urinary microalbuminuria, urinary α1-microglobin, plasma endothelin ,6-Ketoprostaglandin F1α, serum nitric oxide were determined. Result: Compared with model group, the levers of Alb、α1-m、ET-1、TXB2、TXB2/6-keto-PGF1α were markedly reduced in catopril group. Conclusion : It is suggested that catopril could protect vascular Endothelial Function in type 2 diabetes. That might be one of the protective mechanism in diabetic microangiopathy.
