摘要
[目的]探索延龄草的有效繁殖途径。[方法]以延龄草的未成熟种子为外植体,在MS培养基上添加不同的激素配比,在黑暗和光照条件下进行组织培养。[结果]光照条件下,MS+2.0 mg/L6-BA+1.0 mg/L NAA启动延龄草未成熟种子的频率最高,达100.0%;黑暗条件下,MS+2.0 mg/L6-BA+2.0 mg/LIAA启动延龄草的频率最高,达100.0%。光照有利于延龄草未成熟种子启动程度的增加。MS+1.0mg/L6-BA+0.5 mg/LNAA适合诱导延龄草未成熟种子胚性愈伤组织分化和胚状体的发生。显微观察发现诱导出来的延龄草胚状体多处于球形胚和球心胚阶段。[结论]适合延龄草未成熟种子启动的培养基是MS+2.0mg/L6-BA+1.0mg/LIAA。
[Objective] The aim of the research was to explore the effective propagation approach of Trillium tschonoskii Maxim.. [Method] With immature seeds of T. tschonoskii as explants, different hormone combinations were added in MS medium and the tissue culture was conducted under dark and light condition. [Result] Under light condition, MS + 2.0 mg/L 6-BA+ IAA 1.0 mg/L had highest frequency (reaching 100.0 %) of starting the immature seeds of T. tschonoskii. Under clark condition, MS +2.0 mg/L 6-BA +0.5 mg/L IAA had highest frequency (reaching 100.0 %) of starting T. tschonoskii. Light was propitious to the increase of T. tschonoskii start-up degree. MS +1.0 mg/L 6-BA+0.5 mg/L NAA was suitable to induce embryogenic callus differentiation of immature seeds of T. tschonoskii and the generation of embryoid. Micro-observation found that the induced embryoids of T. tschonoskii were mostly at the stages of globular embryo and nucellus embryo. [Conclusion] The suitable medium for the start-up of T. tschonoskii immature seeds was MS+ 2.0 mg/L 6-BA+ 1.0 mg/L IAA.
