摘要
目的探讨CD137分子对单核细胞上CXCR4表达及功能的调节作用。方法采用CD137-Fc融合蛋白刺激单核细胞,流式细胞术(FACS),RT-PCR方法检测CXCR4分子在单核细胞上的表达。Transwell方法分析CD137-Fc激发的单核细胞迁移能力的改变。结果FACS,RT-PCR检测结果显示,CD137-Fc刺激单核细胞48h后,CXCR4蛋白分子及mRNA的表达显著下调。Transwell实验结果显示,CD137-Fc刺激后的单核细胞对趋化因子SDF-1α的趋化能力明显下降(P<0.05)。结论CD137可下调单核细胞上CXCR4表达,进而抑制单核细胞对趋化因子SDF-1α的趋化能力。
Objective To study the regulation of expression and function of CXCR4 by CD137 molecule.Methods The expression of CXCR4 in monocytes was detected by FACS,RT-PCR analysis.Transmigration as-says were performed using a double-chamber transwell system.Results FACS and RT-PCR analysis showed that CD137 could downregulate the expression of CXCR4 on monocytes.Transwell assay revealed that CD137 could inhibit the responses of monocytes to SDF-1α.Conclusion CD137 can suppress SDF-1α-induced migration of monocytes via downregulating the expression of CXCR4.
