甲苯二异氰酸酯对小鼠血液中氧化损伤及能量代谢标志物的影响 被引量:3

摘要 [目的]研究甲苯二异氰酸酯(toluene diisocyanate,简称TDI)对小鼠氧化损伤及能量代谢标志物的影响。[方法]采用50L的静式染毒罐对小鼠进行吸入染毒,每天4h,连续染毒2周。96只昆明种小鼠(雌雄各半)按性别随机分成4组,每组雌、雄各12只。按1/2 LC_(50)、1/4 LC_(50)和1/8 LC_(50)剂量设3个染毒组,另设对照组(正常饲养)。从氧化损伤的角度观察TDI对小鼠血清中的超氧化物歧化酶(SOD)活性、丙二醛(MDA)和谷胱甘肽(GSH)含量的影响;从能量代谢的角度,测定呼吸道接触TDI后,小鼠血液中的琥珀酸脱氢酶(SDH)、乳酸脱氢酶(LDH)及ATP酶(ATPase)活性。[结果]雌、雄各染毒组小鼠血清中MDA含量均呈明显增加(P<0.01);SOD活性和GSH含量与对照组相比均有不同程度的降低。雌、雄各染毒组的SDH活性与对照组相比也均明显增高(P<0.01)。雄性小鼠各染毒组的LDH活性与对照组相比均明显增高(P<0.01),而雌性小鼠只在1/2 LC_(50)浓度下LDH活性才有显著升高(P<0.05)。雌、雄各染毒组小鼠Ca^(2+)-Mg^(2+)-ATPase及Na^+-K^+-ATPase的活性与对照组相比均有下降(P<0.01)。[结论]TDI对氧化损伤和对能量代谢的干扰,导致了染毒动物血液中氧化损伤指标体系和能量代谢指标体系的改变,这些指标可能会成为TDI职业性接触人群早期诊断的参考指标。 [ Objective ] To study the effect on markers of oxidative damage and energy metabolism in mice blood induced by toluene diisoeyanate ( TDI ). [ Methods] The mice were exposed to TDI via respiratory tract in 50 L static inhalation tank. The exposure sustained 4 hours per days for two weeks. The test was divided into three exposed dose groups, namely: 1/2 LC50, 1/4 LC50, and 1/8 LC50 as well as one control group( normal feed ). The effect of TDI on activity of superoxide dismutase( SOD ), contents of malondialdehyde ( MDA ) and glutathione ( GSH ) in mice serum was observed as a measurement for damage due to oxidation. After exposed to TDI, activities of sueeinate dehydrogenase( SDH ), lactate dehydrogenase( LDH )and ATPase in serum were determined with speetrophotometry. [ Results ] The content of MDA increased significantly( P 〈 0.01 ), while the activity of SOD and the content of GSH declined to different degrees in all exposed groups of mice. SDH activity of different group increased significantly, too( P 〈 0.01 ). LDH activity of male mice increased significantly( P 〈 0.01 ), while in female mice only that of 1/2 LC50 group showed the same change ( P 〈 0.05 ). There were notable decreases in activities of Ca^2+-Mg^2+-ATPase and Na^+-K^+- ATPase of different group( P 〈 0.01 ). [ Conclusion ] TDI exerts the changes in target systems of oxidative damage and energy metabolism through mechanisms of oxidative damage and its interference on energy metabolism, those target systems might be the referenced ones of forepart diagnoses for groups vocationally exposed to TDI.
