摘要
[目的]探讨精吡氟禾草灵(Fluazifop-p-butyl)对大鼠的亚慢性毒性,求出最大无作用剂量。[方法]按照GB15670-1995《农药登记毒理学试验方法》中亚慢性90d毒性方法,雌雄性大鼠设低(163、290 mg/kg饲料)、中(653、1160 mg/kg饲料)、高(2610、4640 mg/kg饲料)3个剂量组和1个对照组,测试指标包括血液学、尿液、血液生化检查,病理学检查,每周一次测定体重、进食量,到期解剖测定脏器重量,对结果进行统计学处理。[结果]精吡氟禾草灵高剂量组雌雄性大鼠总摄食量低干对照组,肝/体值、肾/体值高于对照组,雄性大鼠脑/体值高于对照组,脾/体值、睾丸/体值低于对照组;中剂量组雄性大鼠脑/体值、肝/体值、肾/体值高于对照组,脾/体值低于对照组。高剂量组睾丸组织病理学检查可见组织形态学改变。[结论]精吡氟禾草灵可引起SD大鼠睾丸生殖细胞病变;最大无作用剂量雌性为(55.72±4.13)mg/(kg·d),雄性为(22.05±1.56)mg/(kg·d)。
[ Objective ] To study the subchronic toxicity of fluazifop-p-butyl on SD rats and to estimate the NOAEL. [ Methods ] Fluazifop-p-butyl was administered with the diet into female and male Sprague-Dawley rats in three dose groups ( 163, 290 mg/kg -feed, 653, 1 160 mg/kg-feed, 2 610, 4 640 .mg/kg-feed respectively )plus a control group( 0 mg/kg-feed )for 90 days according to China National Standard GB 15670-1995 "Toxicological Test Methods for Pesticides Registration" . Body weight, food consumption, the absolute weight of viscera and the relative weight of viscera, as well as hematology, biochemical indices of the blood and urine, and pathological changes were examined, and then data were treated statistically. [ Results ] A decrease in total food consumption, increase in the organ coefficients of liver, kidney of male and female rats were observed in high dose group, compared with control group. An increase in the organ coefficients of brain and decrease in the organ coefficients of spleen, testis of male rats were observed in high dose group, compared with control group. Nevertheless, an increase of the organ coefficients of brain, liver, kidney and a decrease in the organ coefficients of spleen of male rats were observed in middle dose group, compared with control group. The morphological changes of testes were found in high dose group. [ Conclusions ] Fluazifop-p-butyl could induced the pathological changes in testes, and it's NOAEL were estimated as 55.72±4.13 mg/( kg·d )for female rats and 22.05±1.56 mg/( kg·d )for male rats.
