二价金属离子对博莱霉素作用淋巴细胞SCE的影响

摘要 [目的]观察金属离子和离子络合剂对博莱霉素(BLM)作用淋巴细胞SCE的影响。[方法]在含有一定量BLM的人外周血淋巴细胞培养物中注入一定量的金属离子或离子络合剂,统计淋巴细胞的姐妹染色单体交换(SCE)率。[结果]终浓度为0.5mmol/L和1.0mmol/L的Fe2+、终浓度为2.0mmol/L和4.0mmol/L的Mg2+在BLM切割DNA上有明显的促进作用(P<0.05);终浓度为2.0mmol/L的Fe2+则有极其明显的促进作用(P<0.01);同为2.5mmol/L的EDTA和枸橼酸钠则有明显的拮抗作用(P<0.05)。[结论]一定量的Fe2+和Mg2+的存在对于BLM切割DNA的过程是必需的。 [Purpose ] To investigate the effect of catholyte and its chelating agent on bleomycin treated lymphocyte sister chromatid exchanges (SCE). [Method] SCE was measured after catholytes or chelating agents incubated with cultured human peripheral blood lymphocytes which had been treated with BLM. [Resultsl Both groups of ferrous chloride (0.5 or 1.0mmol/L) and magnesium chloride (2.0 or 4.0mmol/L) showed a significant difference with the control group in cutting lymphocyte DNA fragments (P〈0.05). The group of ferrous chloride (2.0mmol/L) showed a remarkable significant difference with the control group (P〈0.01). Both sodium citrate (SC) and EDTA (2.5mmol/L both) showed a significant difference compared to the control group (P〈0.05). [Conclusions] Both ferrous ion and magnesium ion are necessary to process of bleomycin on lymphcyte DNA fragment cutting.
