反相高效液相色谱法测定活骨胶囊中丹参酮Ⅱ_A含量

Determination of Tanshinone Ⅱ_A in Huogu Capsules by HPLC

目的建立测定活骨胶囊中丹参酮ⅡA含量的反相高效液相色谱法(RP-HPLC法)。方法采用Symmetry C18色谱柱(250mm×4.6mm,5μm),以甲醇-水(80∶20)为流动相,流速为1.0mL/min,检测波长为270nm,柱温为室温。结果丹参酮ⅡA进样量在0.08～0.80μg范围内与峰面积线性关系良好,回归方程Y=4259836.7X-33874.1,r=0.9998,平均加样回收率为99.1%,RSD=1.5%(n=5)。结论RP-HPLC法简便、准确,重现性好,稳定可靠,可用于活骨胶囊的质量控制。

Objective To establish a RP-HPLC method for determination of tanshinone ⅡA in Huogu capsules. Methods A Symmetry C18 （250 mm×4.6mm,5 μm） column was used. The methyl water （80：20） was as the mobile phase. TEe flow rate was 1.0 mL/min. TEe determination wavelength was at 270 nm and the column temperature was maintained at room temperature. Results TEe linear range was 0.08-0.80 μg for tanshinone ⅡA （r=0.9998） and the average recovery rate was 99.1%, RSD=1.5% （n=S）.The linear equation was Y=4 259 836. 7X-33 874. 1. Conclusion The method is simple, accurate, reproducible, reliable. It can be used for the quantitative control of Huogu capsules.
