摘要 目的:探索过氧化酶增殖剂激活受体(PPAR)激动剂在人子宫内膜基质细胞中对于RANTES转录与翻译的作用。方法:Western blot测定PPAR-γ在人子宫内膜基质细胞中的表达;酶联免疫法测定PPAR-γ激动剂对于RANTES蛋白合成的作用;报告基因测定PPAR-γ激动剂对于RANTES转录的作用。结果:①PPAR-γ在子宫内膜基质细胞中表达;②PPAR-γ激动剂罗格列酮显著抑制RANTES的转录65%(P<0.01);③罗格列酮使子宫内膜基质细胞RANTES分泌减少48%(P<0.01)。结论:①子宫内膜基质细胞表达PPAR-γ;②PPAR-γ激动剂抑制子宫内膜基质细胞ANTES的转录和翻译。 Objective: To investigate the effect of Peroxisome Proliferator-Activated Receptor (PPAR-γ) ligands upon transcription and translation of RANTES in human endometrial stromal cells. Methods: Expression of PPAR-γ in endometrial stromal cells was detected by Western blot. The effect of PPAR-γ promoter on production of RANTES by enzyme-linked immunosorbent assay and gene sequencing assay. Results: In cells transfected with RANTES promoter vector, the addition of PPAR-γ ligands inhibited promoter activity by 65% (P〈0. 01). Stromal cells showed a 48% decrease in RANTES protein secretion when treated with a PPAR-γ ligand (P〈0. 01). Conclusion: The use of PPAR-γ ligands to chemokine production and inflammation may be a promising therapy for endometriosis.
