骨康方含药血清诱导大鼠骨髓基质细胞向成骨细胞方向分化的实验研究 被引量:10

摘要 目的:观察骨康方含药血清促进骨髓基质细胞(Marrow Stromal Cells,MSCs)体外向成骨细胞增殖、分化的作用。方法:采用不同浓度的骨康方含药血清联合诱导剂对大鼠MSCs定向诱导分化,观察MSCs诱导前后形态学变化;采用ELISA法检测MSCs骨钙素(Osteocalcin,OC);RT-PCR方法检测MSCsOCN mRNA和Cbfα1mRNA表达。结果:联合使用高、中浓度骨康方加诱导剂能显著提高MSCs增殖和骨钙素的含量,且使Cbfα1 mRNA的表达升高。结论:骨康方具有促进大鼠体外MSCs向成骨细胞增殖和分化的作用,这一作用可能与其升高其Cbfα1 mRNA的表达有关。 Objective: To study the effects of blood-serum containing pharmacon of GuKang on the proliferation and differentiation of bone marrow stromal ceils to osteoblasts in vitro. Methods: The bone marrow stromal cells were cuhured in the media with varied concentrations of blood-serum containing pharmacon of GuKang and inductor. The morphological changes were observed before and after being induced. Osteocalcin was detected by ELISA. OCN mRNA and Cbfα1 mRNA expressions were detected by RT-PCR. Results: High concentration and middle concentrations of GuKang combined with in- ductor all significantly enhanced MSCs proliferation and OC content, and promoted the expression of Cbfα1 mRNA. Conclusion: GuKang could enhance significantly the proliferation and differentiation of rat bone marrow stromal cells in vitro to osteoblasts, which may be mediated by increasing the expression of Cbfα1 mRNA.
